Functional Monitoring of Receptor Tyrosine Kinase Signaling
- Cell-based functional assay for measuring receptor tyrosine kinase (RTK) and signaling. Top panel shows the dose-dependent activation of the endogenous EGF receptor in COS-7 cells. The EC50 value obtained by the RT-CES® system is comparable to the EC50 value obtained by EGFR ELISA assay. Middle panel shows dose-dependent inhibition of EGF-mediated response on the RT-CES® system by an EGFR-specific inhibitor. The IC50 values are comparable to EGFR ELISA assay which was performed side by side.
- Long-term effect of receptor activation on cells (bottom panel). The system allows for monitoring a long term effect of receptor activation in the same cell population. In this example, the cells were treated with EGF. The short term activation of receptor and the long term cell proliferation of the receptor activation can be monitored and recorded.
